چکیده

In this Application Note, separation matrices for protein characterization by capillary gel electrophoresis (CGE) with UV detection from two suppliers, Beckman Coulter and Advanced Analytical, were compared on the Agilent 7100 CE system. Two different sample sets were analyzed: fi rst, a protein size standard and BSA as a test protein for molecular weight determination; second, a reduced antibody standard for quantifi cation of light chain, non-glycosylated heavy chain and heavy chain. To get an estimate for the intermediate precision, these experiments were performed with two different gel lots and capillary batches each. Across these experiments, the gels from both suppliers showed a similar performance. Impurity detection experiments were done with a low molecular weight protein spiked into a non-reduced antibody standard sample. With the gels of both suppliers it was possible to detect this protein down to a level of 0.1 %. Author Christian Wenz Agilent Technologies, Inc. Waldbronn, Germany

Monoclonal Antibody Charge Heterogeneity Analysis by Capillary Isoelectric Focusing on the Agilent 7100 Capillary Electrophoresis System

Due to the importance of monoclonal antibodies (mAb) as therapeutic agents, there is a growing demand for high resolution analytical methods to characterize these complex molecules. One important property of an mAb is its charge state that may change during and after manufacturing. A very useful technique to measure mAb charge heterogeneity is capillary isoelectric focusing (cIEF). This Applica...

Optimization of an rMAb Sample Preparation Scheme for Quantitative CE–SDS Analysis with LIF Detection

www.chromatographyonline.com Electrophoretic techniques, such as sodium dodecyl sulphate polyacrylamide (SDS-PAGE) and isoelectric focusing (IEF) gels, have traditionally been part of release testing in the biopharmaceutical industry. However, these slab gel methods are inconvenient and irreproducible because of the staining/destaining steps used in analyte detection, the use of toxic reagents ...

Method for phosphorothioate antisense DNA sequencing by capillary electrophoresis with UV detection.

The progress of antisense DNA therapy demands development of reliable and convenient methods for sequencing short single-stranded oligonucleotides. A method of phosphorothioate antisense DNA sequencing analysis using UV detection coupled to capillary electrophoresis (CE) has been developed based on a modified chain termination sequencing method. The proposed method reduces the sequencing cost s...
